Amazon rainforest bark extract calms inflammation via nitric oxide pathway

Traditional healers in the Amazon have used breu bark for inflammation for generations, and this study shows the plant chemistry behind that folk remedy actually works through a specific, well-known biological pathway rather than just anecdote.

Breu is a tree from the Brazilian Amazon that local communities have long used to treat swelling and pain. Researchers extracted compounds from its bark, found it's rich in antioxidants like ellagic acid, quercetin, and rutin, and showed that when mice were given the extract, it reduced swelling, pain, and immune cell activity tied to inflammation. The effect worked through a specific chemical signaling chain in the body involving nitric oxide, the same pathway targeted by some existing anti-inflammatory medicines.

At 500 mg/kg, the aqueous bark extract reduced paw swelling by 54%, pain sensitivity by 60%, and neutrophil migration by 74% in mice with carrageenan-induced inflammation.

The extract also cut edema caused by PGE2 (78%), TNF-alpha (72%), and L-arginine (66%), and lowered nitrite levels in peritoneal fluid by 74%.

The anti-inflammatory effect was blocked by ODQ (a guanylate cyclase inhibitor) but not L-NAME, pointing to the L-arginine/NO/cGMP signaling pathway, consistent with in silico binding predictions for PTGS2, NOS, and ALOX5.
