New gene-editing test spots cardamom virus in the field

Cardamom plants can get infected with a virus that causes 'katte disease,' stunting growth and slashing yields, and different regions have their own genetic version of the virus. Researchers built a test using CRISPR gene-editing tools that reads a crushed-up leaf sample and lights up a paper strip if it detects the local Kerala strain of the virus, similar to a rapid COVID test. It works almost as well as expensive lab equipment but takes about an hour and needs no special machinery, so farmers can check plants right where they grow.
Key Findings
The CRISPR-Cas12a lateral flow assay detects the Idukki (cluster IV) strain of cardamom mosaic virus directly from crude plant extracts without RNA purification.
Optimal performance was reached with 10 minutes of RT-RPA amplification, 1 hour of CRISPR reaction time, and specific reporter/Cas12a/crRNA concentrations (500/75/125 nM).
The assay detected as little as 1 femtogram of viral material (240 copies), only 10 times less sensitive than standard qPCR, and was validated on real field samples.
