Blocking a tumor's lactate shuttle may fight cancer growth

Medical Research

This is a human cancer biology review with no connection to plants, gardening, or the outdoors.

Cancer cells produce a lot of lactate as they burn sugar for energy, and two proteins called MCT1 and MCT4 act like doors that let lactate move in and out of tumor cells. This movement helps tumors grow, hide from the immune system, and resist treatment. Researchers reviewed which experimental drugs actually jam these doors shut effectively versus ones that just look promising on paper.

Key Findings

1

MCT1 primarily handles lactate uptake into cells while MCT4 exports lactate from oxygen-starved, glycolytic tumor cells, creating a division of labor within tumors

2

The review distinguishes transporters that are merely present (expressed) from those a tumor actually depends on functionally, a key distinction for drug targeting

3

MCT-targeting drugs are categorized by selectivity (nonselective, MCT1-selective, MCT4-selective, or dual) and evaluated on potency, safety, and clinical readiness

chevron_right Technical Summary

Scientists reviewed how cancer cells use two transport proteins to shuttle lactate (a byproduct of sugar breakdown) in and out of tumors, helping cancer grow, resist treatment, and evade the immune system; the review maps out which drug candidates actually block this process effectively.
