A membrane 'organizer' protein helps plant cells coordinate growth and defense

Plant Signaling

The same molecular sorting system that keeps a plant's immune sensors clustered and ready to fire is likely at work in the tomatoes and peppers in your garden, since these membrane organizer proteins are conserved across flowering plants.

Think of a plant cell's outer membrane as a crowded room where hundreds of different proteins need to find their designated spots to do their jobs, like sensing threats or coordinating growth. Researchers discovered a protein called HIR2 that works like a scaffold, physically gathering certain receptor proteins into clusters so they function correctly. When they removed HIR2 from Arabidopsis plants, the plants grew poorly and had weakened immune responses, showing just how important this organizing role is.

Key Findings

1

HIR2 physically interacts with multiple plasma membrane receptor kinases, including BAK1, BIR2, and BIR3, linking it to BAK1-mediated immune and growth signaling

2

hir2 mutant plants show impaired growth and immunity phenotypes, and advanced microscopy (sptPALM) showed HIR2 directly affects BAK1's mobility and clustering on the membrane

3

AlphaFold 3 modeling and blue-native PAGE experiments show HIR2 forms a multimeric, cup-shaped complex, suggesting it creates structural sub-compartments within the membrane

chevron_right Technical Summary

Scientists found a protein in Arabidopsis plants that acts like an organizer, clustering other proteins together on the cell's outer membrane so they can send signals properly. This helps explain how plant cells coordinate growth and immune responses at a structural level.

The Arabidopsis SPFH protein HIR2 modulates receptor signaling and plasma membrane organization.

Plant plasma membrane proteins are organized into distinct domains; however, the principles that govern this spatial organization are not fully understood. HYPERSENSITIVE-INDUCED REACTION (HIR) pro...
