New soil method reveals bacteria hitchhiking on fungal threads

The mushroom threads spreading invisibly under your garden beds carry their own hidden bacterial passengers, and this new extraction trick means scientists can finally study those soil partnerships without contaminating the sample with sand.
Underground, fungi form vast networks of microscopic threads called hyphae, and tiny bacteria cling to those threads like passengers on a subway line. Pulling those threads out of soil to study them has always been messy and slow, often requiring sand or glass beads that mess up the results. This new technique uses sieving and a sugar-water spin cycle to gently separate the threads from soil, keeping their bacterial hitchhikers intact and cutting the lab work time by more than half.
Key Findings
The sieving and sucrose centrifugation (SSC) method processes soil samples up to 2.5 times faster than earlier manual protocols.
SSC yielded the highest hyphal length density (HLD) among all methods tested, without requiring sand or glass bead additives.
The technique preserves surface-attached bacteria on fungal hyphae, enabling targeted microbiome analysis via microscopy, molecular tools, and next-generation sequencing.
